Substrate concentration
With a constant enzyme concentration, the enzyme action increases with increasing substrate concentration until a certain level when all active sites of enzymes are occupied and the rate of reaction then levels off. Before the graph levels off, substrate concentration is the limiting factor. When the graph levels off, enzyme concentration becomes the limiting factor.
